Diaporthe tectonendophytica

Diaporthe tectonendophytica isolate MFLUCC 13–0471 (ex-type) grouped closest to, but distinct from, D. longicolla (FAU 599) and D. unshiuensis (ZJUD 52) with 51 % MPBS and 99 % MLBS. Comparison of the polymorphisms of ITS, TEF1, TUB and CAL between D. tectonendophytica and its closest relatives D. longicolla (FAU 599) and D. unshiuensis (ZJUD 52) showed D. longicolla with 15, 33, 31 and 31 polymorphisms, D. unshiuensiswith. There is no CAL sequence data of D. unshiuensis thus, the comparison could not be made. Based on multiple gene phylogenetic and polymorphisms nucleotide differences, it is suggested that Diaporthe tectonendophytica is a novel species.

Seven isolates of D. tectonae (MFLUCC 12–0777, MFLUCC 12–0782, MFLUCC 13–0460, MFLUCC 13–0476, MFLUCC 14–1132, MFLUCC 14–1134 and MFLUCC14–1139) placed together in 93 % MPBS and 92 % MLBS and these seven isolates placed within other isolates of D. tectonae (MFLUCC 14–1135 and MFLUCC 14–1138) with 98 % MPBS and 98 % MLBS. All nine isolates group close to, but separate from D. tulliens is BRIP 62248a with 100% MPB Sand 100 % MLBS. Diaporthe tectonae isolate MFLUCC 12–0777 (ex-type) showed multiple nucleotide differences from D. tulliensis BRIP 62248a (ex-type) for the ITS, TEF1 and TUB regions analysed with 7, 12 and 5 polymorphisms respectively. There is no CAL sequence data of D.tulliens is BRIP 62248a thus comparison could not be made, but it is suggested that our collections are a new species.

Diaporthe tectonigena (MFLUCC 12–0767) grouped next to but separate from Diaporthe sp. (MFLUCC 12–0767) with support 89 % MPBS and 97 % MLBS, suggesting that it represents a novel species.

 

There are four novel species based on the combination of ITS, TEF1, TUB and CAL phylogeny and polymorphic nucleotide comparisons including D. neoraonikayaporum (MFLUCC 14–1133, MFLUCC 14–1137 and MFLUCC14–1136), D. tectonendophytica (MFLUCC 13–0471), D. tectonae (MFLUCC 12–0777, MFLUCC 12–0782, MFLUCC 13–0460, MFLUCC 13–0476, MFLUCC 14–1132, MFLUCC 14–1134, MFLUCC 14–1135, MFLUCC14–1138 and MFLUCC 14–1139) and Diaporthe tectonigena (MFLUCC 12–0767).

Notes: Diaporthe tectonendophytica has been isolated as an endophyte on teak. In comparison to phylogenetically close species, the alpha conidia of D. tectonendophytica are ellipsoid, but ellipsoid to ovoid for D. longicolla whilst they are ellipsoidal or clavate in D. unshiuensis. Alpha conidia of D. tectonendophytica are shorter and wider than D. longicolla, shorter and narrower than D. unshiuensis. The multi-gene phylogeny and polymorphic nucleotides comparisons distinguish D. tectonendophytica from D. longicolla and D. unshiuensis.
